1928年4月，陈寿昌被抽调到中央情报科，成为周恩来领导的中央特科“三陈”之一，主要负责情报和交通联络工作。他与“龙潭三杰”单线联系，及时得到的机密情报。同时，他广泛搜集很多中英文报纸，细致分析不同来源的资料，准确判断敌人动向，为开展秘密斗争确定对策，为中央工作决策提供参考。

上海街头经常发生“抄靶子”，即军警突然搜查行人。陈寿昌带着机密情报和文件，屡屡碰到军警“抄靶子”，由于准备充分，伪装巧妙，往往能化险为夷。他有时穿一件旧灰布长衫，戴一副近视眼镜，扮作小学教员模样，有时又是一身专做西式服装的“红帮裁缝”打扮。有一次，陈寿昌在途中又被军警拦住盘问。他用宁波方言与军警交谈。对方听他说的是宁波话，手里又夹着裁缝布包，打开一看里面全是剪裁衣服用的尺子、剪刀、划粉和各种样式的布料，还真以为遇到了“红帮裁缝”，很快放行。
